Microbiology - FAQs

1) Method SM 9230 B (MPN) for fecal strep and enterococci analysis states to use PSE agar to for the confirmation test but PSE agar is no longer available. So what do we use? Answer: BEA is the standard substitute for PSE. There is a slight difference in the buffering reagents used and a little less azide.

2) The temperature of microbiological samples arriving at the lab is important but how can this be determined without contaminating the sample by using a standard thermometer? Answer: Infrared thermometers with laser sighting can be used. EPA has used them without worrying about the exposure of the organisms to the laser because of the short exposure time. However, these thermometers, as with traditional thermometers, need to be calibrated annually for accuracy.
